Kaskaskia was the first major town in Illinois (and the first state capital), peaking about 1815 somewhere around 5,000 people. However it shrank quickly due to losing its capital status in 1819 and frequent flooding; by 1820 it had fallen below the 2,500 cutoff to be considered urban, and was practically abandoned by 1840.

Kaskaskia is now the only part of Illinois that is west of the Mississippi River as a result of flooding in the 1880s (not the New Madrid earthquake)

Nauvoo is reported to have had a population of 12,000 in 1844 and was the state's largest city, but its standing was short-lived as most of the Mormons left for Utah before the 1850 census.

In 1850, it was down to 1,130, and unfortunately the 1840 Census isn't online, so I've had to find its numbers reproduced in later Censuses; Nauvoo's has not been found, although the first Mormons didn't arrive until late 1839, implying a very small population in the spring of 1840. Kaskaskia by 1850 had plunged to 513 people.
